DNA Microarray and insitu analysis of gene expression in Drosophila embryos.
Microarray analysis : Wild type embryos were collected in 1 hour intervals spanning the first 12 hours of Drosophila
embryogenesis. Each collection was repeated three times independently. Total RNA was isolated using Ambion's RNAwiz and labelled
using standard Affymetrix Gene Chip protocols and hybridised to Drosophila Gene Chip arrays.
Data were analysed by Affmetrix Microarray Suite 4.0 and dChip algoritm.
RNA insitu hybridisation : RNA insitu hybridisation in 96 well format was performed as described (link).
Embryos collections were biased towards early stages of embryogenesis when gap and pair-rule patterning processes occur. Images
were recorded using Zeiss Axiophot light microscope and stored in a mySQL database.
